Icilin is a cooling agent that activates the novel cold receptors TRPM8 (CMR1) and TRPA1 (ANKTM1/TRPN1), members of the TRP ion channel family. Induces currents in CMR1-expressing HEK 293 cells (EC50 = 0.36 μM) more potently than menthol or low temperatures. Produces "wet shakes" in vivo.
Icilin, also known as AG-3-5, is a potent super-agonist that targets the transient receptor potential M8 (TRPM8) ion channel. It demonstrates dose-dependent activation of TRPM8 in the presence of EGTA, with an effective concentration (EC50) of 1.4 μM. Recognized as a "super-cooling agent," Icilin plays a role in attenuating autoimmune neuroinflammation by influencing the T-cell response.
