DAB substrate kit (ab64238) is used to detect horseradish peroxidase (HRP)/peroxidase enzyme in immunohistochemistry (IHC) staining. Produces a crisp brown stain that is alcohol-insoluble. Part of our Lab essentials imaging workflow offering. - Proven performance: cited in over 480 publications - Working solution stable for up to 6 hours after mixing - Refrigerator-stable for extended usability
The DAB substrate kit (ab64238) working reagent is stable for up to 6 hours after mixing the two components.
DAB (3,3'Diaminobenzidine) is the most commonly used chromogen for immunohistochemical staining. In the presence of horseradish peroxidase (HRP) / peroxidase, DAB produces a brown precipitate that is insoluble in alcohol. This product is a two component form consisting of a liquid, refrigerator stable DAB Chromogen and DAB Substrate.
Staining Protocol (see protocol booklet for further details):
Equilibrate all materials and prepared reagents to room temperature prior to use
1. Add 30 μl (1 drop) DAB Chromogen to 1.5 ml (50 drops) of DAB Substrate, mix by swirling and apply to tissue.
2. Incubate for 1-10 minutes. Rinse 4 times in buffer.
3. Apply counterstain according to manufacturer’s instructions.
4. Dehydrate if required and coverslip.
